Povzetek

V raziskavi smo analizirali strukturo in ultrastrukturo epitelnih celic Malpighijevih cevk (MC) rjave jamske kobilice (Troglophilus cavicola) v obdobju prezimovanja. Vrsto uvrščamo med troglofile, kar pomeni, da je prilagojena tako na površinske kot na podzemeljske habitate, kjer prezimuje. Namen raziskave je bil proučiti spremembe v strukturi in ultrastrukturi epitelnih celic MC, vključno s prisotnostjo avtofagnih struktur in njihovo potencialno funkcijo med prezimovanjem. Prezimovanje je obdobje naravnega stradanja rjave jamske kobilice in je zato primeren del življenjskega cikla, ko lahko proučujemo posledice stradanja na strukturo celice. Kobilice obeh spolov smo nabrali v treh časovnih obdobjih (november, januar, marec). S pregledom vsebine prebavila smo potrdili, da se osebki v času prezimovanja ne prehranjujejo. Strukturo in ultrastrukturo epitelnih celic MC smo opazovali s pomočjo svetlobnega in transmisijskega elektronskega mikroskopa (TEM) v vseh treh obdobjih. Razlik v zgradbi epitela MC med samci in samicami nismo opazili v nobenem obdobju. Pred prezimovanjem, v novembru, v epitelnih celicah MC nismo opazili avtofagnih struktur. Te so bile prisotne v celicah na sredini (januar) in ob koncu prezimovanja (marec). Na tej osnovi lahko sklepamo, da v času prezimovanja kobilice z avtofagnimi procesi vzdržujejo homeostazo celic z njihovo oskrbo s hranili. V epitelnih celicah MC T. cavicola smo med prezimovanjem opazili tudi številne sferite. Njihova zgradba se je med prezimovanjem spreminjala. Posamezne plasti materiala so se postopoma uporabile in ob koncu prezimovanja so bili sferiti večinoma izpraznjeni. Z raziskavo smo potrdil domnevo, da se v epitelnih celicah MC med prezimovanjem sproščajo mineralne snovi iz sferitov in iz avtofagnih struktur.

Sekundarni povzetek: In this research we studied the structure and ultrastructure of the epithelial cells of Malphigian tubules (MTs) in the cave cricket Troglophilus cavicola during overwintering. This species is classified as troglophile, as it is adapted for living in both the epigean and the hypogean habitats. The aim of the study was to investigate changes of structure and ultrastructure of MTs and to evidence the presence of autophagic structures to estimate its potential role during overwintering. Winter dormancy is a natural starvation period, providing the opportunity to study consequences on cell structure during starvation. Male and female specimens were collected in three times frames (November, January and March). By the gut investigation we confirmed that the cave crickets did not feed during overwintering. The structure and ultrastructure of the MT epithelial cell were studied by light microscopy and transmission electron microscopy (TEM). There were, generally, no differences in the structure of epithelium of MTs between both sexes. At the beginning of overwintering, in November, no autophagic structures were present in MTs epithelial cells, while these were present in the middle (January) and at the end of the overwintering (March). We conclude that in T. cavicola, autophagy is a response to starvation supporting homeostatic processes during winter dormancy. During overwintering, numerous spherites were present in MTs epithelial cells. Their structure considerably modified in that period. Spherite layers have been gradually disintegrated during overwintering, resulting in mostly empty spherites at the end of overwintering. With this research we confirmed our hypothesis that during overwintering materials from both spherites and autophagic structures are released in MTs epithelium cells in order to maintain their basal metabolism.
