Abstract
Hrenova peroksidaza je encim, ki ga najdemo v koreninah hrena in so jo preučevali več kot stoletje. S pomočjo njene tridimenzionalne strukture so bile ugotovljene njene uporabne lastnosti. Zlasti je pomembna za farmacevtsko in biotehnološko industrijo, saj je zanimanje za biokatalizatorje v kemijski proizvodnji vedno večje. Industrijsko je pomembna tudi imobilizacija encimov, saj ta zvišuje stabilnost in omogoča ponovno uporabo encima. Ena izmed teh metod je CLEAs, zamreženi encimski skupki, ki predstavlja imobilizacijo brez nosilca. Gre za zamreženje encima. Če takšne skupke hrenove peroksidaze izpostavimo različnim pogojem, kot sta tlak in temperatura, v različnih časovnih intervalih, ugotovimo, da s časom aktivnost agregatov upada.

Secondary abstract: |
Horseradish peroxidase is an enzyme that could be found in horseradish roots. It has been studied for more than a century. The threedimensional structure of peroxidase revealed a lot of advantages. Because of the interests in biocatalyst, peroxidase is very important in pharmaceutical and biotech industry where immobilization of enzymes is also important. Immobilization increases stability of enzymes and allows re-use. One of immobilization methods is CLEA, cross-linked enzyme aggregates. This method presents carrier-free immobilized enzymes where enzymes are cross-linked. If aggregates of HRP are exposed to different conditions such as pressure and temperature, in different intervals, a decrease in activity of aggregates was detected. |
