Abstract

Namen diplomskega dela je bil določiti vpliv superkritičnega ogljikovega dioksida (SC CO2) na aktivnost zamreženih encimskih agregatov (CLEAs) iz encima celulaze. CLEAs in prosti encim celulaze smo v visokotlačnem šaržnem reaktorju izpostavili SC CO2 pri temperaturi 50°C in tlaku 200 bar. Istočasno smo CLEAs in prosto celulazo inkubirali pri atmosferskem tlaku in 50°C. Iz rezultatov je razvidno, da se je aktivnost CLEAs s podaljševanjem časa izpostavljanja v SC CO2 zviševala medtem, ko se je aktivnost pri izpostavitvi CLEAs v atmosferskemu tlaku znižala. Določili smo tudi koncentracijo proteinov CLEAs in proste celulaze. S podaljševanjem časa izpostavitve v SC CO2 se je koncentracija proteinov zvišala. Optimirali smo različne parametre pri pripravi CLEAs iz celulaze. Najvišja aktivnost CLEAs je bila dosežena, ko smo albumin razredčili v PBS pufru s koncentracijo 0,02 M.

Secondary abstract: The purpuse of the diploma thesis is to examine the effect of supercritical carbon dioxide (SC CO2) on the activity of cross-linked enzyme aggregates from cellulase. CLEAs and pure enzyme cellulase were exposed to SC CO2 in high pressure batch reactor at 50 °C and 200 bar. At the same time CLEAs and free cellulase were exposed to atmospheric pressure at 50 °C. The results showed that activity of CLEAs in dependance of incubation time in SC CO2 increased, meanwhile the activity of CLEAs in atm. pressure decreased. The concentration of proteins in CLEAs and free cellulase was also measured. In prolongation of incubation time in SC CO2 the concentration of proteins in CLEAs and free cellulase increased. Different parameters was optimized in preparation of CLEAs from cellulase. The highest activity of CLEAs was obtained when the albumin was diluted in PBS buffer with the concentration 0,02 M.
