Abstract

V tej raziskavi smo razvili novo metodo imobilizacije β-galaktozidaze na zvitke steklene tkanine. Raziskali smo uporabnost steklene tkanine kot vlaknastega nosilca z visoko poroznostjo in odprto strukturo. Nov postopek zagotavlja preprosto, cenovno ugodno in industrijsko primerno metodo imobilizacije. Glutaraldehid smo uporabili kot nespecifično povezovalno sredstvo za imobilizacijo β-galaktozidaze na stekleno tkanino. Le-ta je omogočal kovalentno vezavo encima na modificiran nosilec. Učinkovitost imobilizacije encima smo testirali na dobro poznani reakciji hidrolize laktoze. Vse eksperimente smo izvajali v laboratorijskem kontinuirnem reaktorju. Preučevali smo vpliv temperature,  = (20, 25 in 30) °C, pretoka, qV = (1, 2 in 3) mL/min in pH = (6, 7 in 8) na stabilnost in aktivnost β-galaktozidaze med hidrolizo. Presnovo laktoze smo določali z merjenjem koncentracije glukoze. Dosegli smo visoko učinkovitost imobilizacije in končno aktivnost encima. Z večkratno zaporedno uporabo istega imobiliziranega encima je ostala presnova praktično nespremenjena, kar kaže na možno uporabo pri industrijskih aplikacijah.

Secondary title: Immobilization of beta galactosidase on glass cloth rolls - efficiency study
Secondary abstract: In this study we developed a method for β-galactosidase immobilization using special glass cloth rolls as a new designed support. The effectiveness of glass cloth as a fibrous matrix with high porosity and open structure was explored. The new procedure provides simple, inexpensive and industrially applicable method and also support for active and stable immobilized biocatalyst. Glutaraldehyde was used as a non-specific cross-linking agent for fixing the β-galactosidase on the glass cloth. Enzyme was covalently bonded on modified support. We tasted the efficiency- of bonding on well-known hydrolysis of lactose. All experiments were carried out in the laboratory continuous reactor. The influence of reaction temperature (20, 25 and 30 °C), substrate flow rate (1, 2 and 3 mL/min) and pH (6, 7, 8) on reaction conversion were studied. Reaction efficiency was monitored by measuring the glucose concentration using spectrophotometer. We reached high immobilization efficiency and final enzyme activity. The optimum reaction temperature, substrate flow rate and pH were found. β-galactosidase immobilized on glass cloth rolls was successfully used in continuous biocatalyst reactor for hydrolysis of lactose. Activity and stability during the reuse of enzyme entrapped on the glass cloth roll remained almost unchanged, showing high perspective for possible industrial applications.
