Abstract

Namen diplomskega dela je bil raziskati antioksidativno in protimikrobno učinkovitost ekstraktov cejlonskega cimeta (Cinnamomum zeylanicum) s ciljem odkritja naravne alternative sintetično proizvedenim zdravilom oz. prehranskim dopolnilom. Z uporabo konvencionalnih ekstrakcijskih metod in različnih topil smo pridobili ekstrakte cejlonskega cimeta, pri čemer nas je zanimal izkoristek ekstrakcije. Sledile so spektrofotometrične analize. Določili smo antioksidativni potencial z radikalsko metodo DPPH, vsebnost totalnih fenolov in proantocianidinov. Protimikrobno učinkovitost smo preverili v razmerju z grampozitivno bakterijo Staphylococcus aureus, gramnegativno bakterijo E. coli ter kvasovko Candida albicans. Uporabili smo difuzijsko metodo z diski, izvedena pa je bila tudi analiza ekstraktov s HPLC (tekočinska kromatografija visoke ločljivosti), s katero smo določevali prisotnost desetih fenolnih spojin. Rezultati kažejo visoko antioksidativno aktivnost alkoholnih ekstraktov. Za izolacijo komponent z antioksidativnimi lastnostmi se je kot najprimernejše topilo izkazal metanol, kot učinkovita ekstrakcijska metoda pa ekstrakcija po Soxhletu, kjer smo dobili najvišje izkoristke ekstrakcij ter visoke vsebnosti proantocianidinov. Kljub temu pa je zaradi prisotnosti visokih temperatur najverjetneje prišlo do degradacije nekaterih komponent, saj pri ekstrakciji s hladnim topilom namreč dobimo ekstrakte z višjo vsebnostjo fenolnih spojin ter višjo antioksidativno učinkovitostjo. Etanolni ekstrakti so se izkazali za učinkovite pri zaviranju rasti kvasovke Candida albicans, vodni in etanolni ekstrakti pa uspešno delujejo proti bakteriji Staphylococcus aureus. Rezultati so ovrednoteni s kvalitativno in kvantitativno analizo fenolnih spojin v ekstraktih.

Secondary abstract: The aim of the diploma thesis was to investigate antioxidant and antimicrobial effects of Ceylon cinnamon extracts (Cinnamomum zeylanicum) in order to discover a natural alternative to synthetically produced drugs and food supplements. The cinnamon extracts were obtained by using conventional extraction methods and various solvents, with the aim to achieve the highest extraction efficiency. A spectrophotometric analysis followed, where the antioxidant potential was determined by using the radical DPPH, the total phenol content and the proanthocyanidin content methods. The antimicrobal activity was investigated against Gram-positive bacteria Staphylococcus aureus, Gram-negative bacteria E. coli and yeast Candida albicans with a disc diffusion method. The analysis of extracts with HPLC (high-performance liquid chromatography) was then performed in order to determine the presence of ten phenolic compounds. The results have shown a high antioxidative activity of alcohol extracts. Methanol has proved to be the most suitable solvent for the isolation of components with antioxidant properties, while the Soxhlet extraction has yielded the highest extraction efficiency and content of proanthocyanidins. Nevertheless, due to the high temperatures, most of the components have been degraded, as extracts, made by extraction with cold solvent, contain higher amount of phenolic compounds and antioxidant efficiency. Ethanolic extracts have proven to be effective in inhibiting the growth of yeast Candida albicans, while ethanolic and aqueous extracts have worked well against Staphylococcus aureus. The results have been evaluated by qualitative and quantitative analysis of phenolic compounds in extracts.
