Povzetek
Namen diplomskega dela je raziskati antioksidativne lastnosti ekstraktov jagod črnega bezga (Sambucus nigra) s ciljem ovrednotenja izbranega naravnega materiala kot alternative sintetičnim zdravilnim učinkovinam. Uporabili smo tri različne konvencionalne ekstrakcijske metode in dve različni topili, da smo pridobili šest različnih ekstraktov jagod črnega bezga. Po računanju izkoristka ekstrakcij so sledile spektrofotometrične analize. Določali smo vsebnosti totalnih fenolov, proantocianidinov in karotenodiov, ki so znani po antioksidativnih lastnostih. To lastnost ima tudi vitamin C, katerega maso smo določali s titracijo. Na koncu je sledila analiza ekstraktov s tekočinsko kromatografijo z masno spektrometrijo (LC-MS), s katero smo določevali prisotnost osmih fenolnih spojin. Rezultate smo torej dobili s kvantitativno in kvalitativno analizo ekstraktov.
Rezultati kažejo, da so koncentracije vseh osmih fenolnih spojin najvišje pri ekstrakciji po Soxhletu, kjer smo za topilo uporabili mešanico etanola in vode (etanol : voda = 60 : 40 vol. %). Izkoristki so pri nekaterih drugih ekstrakcijah višji, s tem tudi večja vsebnost totalnih fenolov in proantocianidinov, vendar pa je tam koncentracija teh osmih fenolnih spojin nižja. Karotenoidi so prisotni v vseh ekstraktih, vendar v manjših količinah. Ugotovili smo tudi prisotnost vitamina C, kar smo dodatno potrdili z visokimi koncentracijami rutina, fenolne spojine, ki vpliva na delovanje tega vitamina.

The aim of this diploma thesis is to investigate the antioxidant properties of black elderberry extracts (Sambucus nigra) in order to evaluate selected natural compounds as an alternative to the use of synthetic drugs. Three different conventional extraction methods and two different solvents were used, resulting in six different extracts of black elderberry. After calculating the extraction yields, spectrophotometric analyses were performed. The content of total phenols, proantocyanidins and carotenoids, known for their antioxidant properties, was determined. Vitamin C, whose mass was determined by titration, is also known for its antioxidant activity. Finally, the extracts were analysed by liquid chromatography with mass spectrometry (LC MS) to determine the presence and amount of eight phenolic compounds. Thus, the LC MS allowed a quantitative and qualitative analysis of the extracts.
The results show that the concentrations of the eight phenolic compounds measured are highest in the case of Soxhlet extraction, in which a mixture of ethanol and water was used as solvent (ethanol : water = 60 : 40 vol. %). For the other extracts, the yield is higher, as well as the content of total phenols and proanthocyanidins, but the concentration of the selected eight phenolic compounds is lower. Carotenoids are present in all extracts, but in lower amounts. The presence of vitamin C in the extracts was also confirmed, which is in agreement with the results of LC-MS showing high concentrations of rutin, a phenolic compound that affects the action of this vitamin. |
