Abstract

Aloja, rastlina nesmrtnosti, kot jo nekateri poimenujejo, je v današnjem času izredno priljubljena, predvsem zaradi svojih zdravilnih učinkov. Prihaja iz družine zlatokorenovk. Poznamo več kot 200 vrst aloje, vendar se v zdravilne namene uporabljata samo dve, to sta Aloe arborescens in Aloe barbadensis, ki jo imenujemo tudi Aloe vera. Aloja je sestavljena iz 99 % vode, ostali 1 % pa predstavljajo zdravilne učinkovine, kot so vitamini, encimi, sladkorji, minerali, saponini, lignin, salicilna kislina in aminokisline. V diplomski nalogi bomo podrobneje opisali encime, ki jih vsebuje aloja, kot tudi določili njihove vsebnosti v hladilnem gelu, ki je v zadnjem času izredno priljubljen pri celjenju ran. V okviru diplomskega dela smo pripravili lasten gel iz aloje in primerjali encimsko aktivnost med kupljenim in domačim gelom. Encimi, ki jih vsebuje aloja, so amilaza, bradikinaza, karboksipeptidaza, katalaza, celulaza, kreatin-foskinaza, fosfataza, lipaza, proteaza, transminaza in tirozinaza. Odločili smo se, da bomo preverili aktivnosti naslednjih encimov: celulaze, α-amilaze, proteaze, peroksidaze, katalaze, transglutaminaze in lipoproteinske lipaze. Opravili smo encimske teste, izmerili absorbanco na UV/Vis-spektrofotometru v pripravljenih vzorcih ter s pomočjo izmerjene absorbance določili aktivnost encimov v A. arborescens in A. barbadensis. Aktivnosti encimov so bile nizke zaradi velikega deleža vode v aloji.

Secondary title: Study of the activity of certain enzymes in the gel of aloa vera
Secondary abstract: Aloe or the plant of immortality as some would call it, is very popular nowadays, mainly because of its healing effects. It originates from the family of Asphodelaceae. There is a lot of species of Aloja, over 200 of them, but only 2 are used for its healing effects, and those are A.arborescens and A. barbadesis which is more known as Aloe vera. Aloe is composed from 99% of water and the remaining 1% are active substances such as vitamins, enzymes, sugars, minerals, saponins, lignin, salicylic acid and amino acids. In this graduation thesis we will mainly focus on enzymes that aloe consists of and also the presence of enzymes in cooling gel, which is very popular lately for its healing properties for different wounds. We therefore decided to make our homemade aloe cooling gel and then compare its enzyme activity to the one bought in the store. Enzymes that aloe contains of are amylase, bradykinase, carboxypeptidase, catalase, cellulase, creatine phoskinase, phosphatase, lipase, protease, transminosis and tyrosinase. We decided to determine the activity of the following enzymes: cellulase, α-amylase, protease, horseradish peroxidase, catalase, transglutaminase, and lipoprotein lipase. Some tests, measurements were performed and the absorbance on the UV-Vis spectrophotometre was measured, which helped us to determine the activity of enzymes. The activites were very low, due to large amount of water in Aloe.
