Abstract

Namen diplomskega dela je bil določiti vsebnost izbranih kovin v koreninah medvrstnih križancev bezga. Pred izvedbo analiz na ICP/OES je bilo treba trdne vzorce pretvoriti v tekočo obliko, ki je primerna za analizo. To sem storil s kislinskimi razklopi vzorcev. Kislinski razklop vzorcev sem izvajal ob prisotnosti HNO3, H2SO4 in H2O2. Tako pripravljeni vzorci so bili razredčeni z 1% HNO3 in shranjeni v 50-mL viale. Sledila je analiza z ICP/OES in za primer magnezija tudi AAS. Pred začetkom merjenja kovin v vzorcih sem izvedel umeritveno krivuljo z uporabo ICP-večelementne standardne raztopine VI (Merck). Vzorcem sem iz umeritvenih krivulj določil koncentracije kovin, v primeru magnezija in kalija pa sem moral vzorce tudi razredčiti. Koncentracije kovin sem interpretiral in določil linearnost merilnega območja.

Secondary abstract: The purpose of my thesis was to determine concentrations of certain metals in the roots of interconnected crosses of elderberry. Before the ICP/OES analysis, I had to convert solid samples into a liquid form suitable for analysis. I have done this using the acid digestion technique. For this technique I used HNO3, H2SO4 and H2O2. The samples thus prepared were diluted with 1 % HNO3 and were kept in 50 mL vials. The samples were ready for the ICP/OES analysis and in the case of magnesium the AAS analysis was used too. Before the analysis of the samples of elderberry roots I performed the calibration, using a multistandard solution VI (Merck). I have determined concentrations of metals in samples from the calibration curve. For magnesium and potassium, the dilution was needed. I have interpreted the concentrations of metals and determined the linearity of the measuring zone.
