Abstract

Zaradi daljšanja življenjske dobe prevalenca nevrodegenerativnih bolezni narašča, patogeneza večine teh bolezni pa ostaja nejasna. Napredek na področju tehnologij preurejanja genomov, kot je tehnologija CRISPR/Cas, nam omogoča pripravo celičnih linij in živalskih modelov, s katerimi lahko modeliramo genetske napake, povezane z nevrodegenerativnimi boleznimi pri človeku. Diplomska naloga podaja aktualen pregled dosedanjih raziskav na področju nevrodegenerativnih bolezni in opisuje potencial tehnologije CRISPR/Cas za pripravo celičnih in živalskih modelov. Živalski in celični modeli, ki posnemajo patologijo človeških nevrodegenerativnih bolezni so ključni za pridobivanje pomembnih informacij o poteku bolezni in možnih načinih zdravljenja ali upočasnitve napredovanja bolezni.

Secondary title: Using CRISPR/Cas technology for modeling neurodegenerative diseases
Secondary abstract: Due to the increase in life expectancy, the prevalence of neurodegenerative diseases is increasing, however the pathogenesis of most of the neurodegenerative diseases remains unclear. Advances in gene editing technology, such as CRISPR/Cas, allows us to create cellular and animal models for neurodegenerative diseases in humans. This thesis gives an up-to-date overview of the already conducted research on neurodegenerative diseases and discusses the potential of CRISPR/Cas technology for the design of cellular and animal models that replicate pathology of human neurodegenerative diseases. Animal models are the key to obtaining important information about the course of the neurodegenerative diseases, possible treatments or the process of slowing down the progression of the disease.
