Abstract
Namen diplomske naloge je ugotoviti, kako način in čas ekstrahiranja ter topilo vplivajo na končne ekstrakte, pridobljene iz posušenih listov južnoameriške rastline Yerba mate (Ilex paraguariensis). Izvedli smo ekstrakcijo po Soxhletu in superkritično ekstrakcijo s CO2 ob dodatku različnih topil in z različnimi časi ekstrahiranja. Ker je rastlina znana po zdravilnih in tudi poživljajočih učinkih, smo v ekstraktih analizirali dvoje; antioksidativno aktivnost ter vsebnost petih učinkovin; kofeina, teobromina, teofilina, kofeinske ter klorogenske kisline. Za določitev antioksidativnosti smo uporabili radikalsko metodo DPPH, za določitev učinkovin pa metodo tekočinske kromatografije z masno spektrometrijo. Želeli smo določiti optimalne pogoje ekstrakcije.
Rezultati kažejo, da na kvaliteto ekstraktov vplivajo ekstrakcijska metoda, topilo ter čas ekstrakcije. Ugotovili smo, da je mešanica etanola in vode primernejše topilo kot etanol. Pri ekstrakcijah s tem topilom smo dobili višje izkoristke ter več ekstrahiranih aktivnih učinkovin, antioksidativna aktivnost pa je primerljiva. Najslabše rezultate kaže superkritična ekstrakcija s CO2, s kotopilom etanolom.

Influence of extraction procedure on the quality of Yerba mate (ilex paraguariensis) extract |

The purpose of this thesis is to determine how the method, time of extraction and solvent affect the final extracts obtained from the dried leaves of the South American plant Yerba mate (Ilex paraguariensis). Soxhlet extraction and supercritical extraction with different solvents and different extraction times were performed. Since the plant is known for its healing as well as invigorating effects, we analyzed two things; antioxidant activity and the content of five active ingredients; caffeine, theobromine, theophylline, caffeic and chlorogenic acids. The radical DPPH method was used to determine antioxidant properties, and the liquid chromatography method by mass spectrometry was used to determine the active ingredients. We wanted to determine the optimal extraction conditions.
The results show that the quality of the extracts is influenced by the extraction method, solvent and extraction time. We found that a mixture of ethanol and water is a better solvent than ethanol. Extractions with this solvent gave higher yields and higher content of extracted active ingredients. The antioxidant activity was comparable. Supercritical extraction with CO2 and ethanol was performing the worst. |
