Abstract

Diplomsko delo zajema študijo stabilnosti encima lakaze v neraztopljeni obliki (praškasti obliki) in raztopljenega v pufru v superkritičnem ogljikovem dioksidu (SC CO2). Pri tem smo uporabljali šaržni reaktor, v katerem je bil encim izpostavljen SC CO2 pri različnih pogojih. Proučili smo kakšen vpliv ima izpostavitev obeh oblik lakaz v SC CO2 pri različnih temperaturah in tlakih. Ugotovili smo, da na stabilnost lakaze v SC CO2 vpliva tako tlak kot temperatura. Večji vpliv na preostalo aktivnost ima temperatura, saj smo pri višji temperaturi izpostavitve vzorcev v SC CO2 zasledili večji upad v preostali aktivnosti neglede na izbrani tlak. Izkazalo se je, da je lakaza v praškasti obliki bolj stabilna v SC CO2 kot lakaza raztopljena v pufru.

Secondary title: Stability of laccase in SC CO 2
Secondary abstract: The diploma presents a study of the stability of the enzyme laccase in undissolved form (powder form) and dissolved in buffer in supercritical carbon dioxide (SC CO2). The batch reactor in which the enzyme was exposed to SC CO2 under different conditions was used. The effect of exposure to both forms of laccase in SC CO2 at different temperatures and pressures was studied. We found that both pressure and temperature influence the stability of laccase in SC CO2. Temperature has a greater influence on the residual activity, since the activity of the samples in SC CO2 was lower at higher temperature, regardless of pressure. Laccase in powder form has been shown to be more stable in SC CO2 than laccase dissolved in buffer.
