Abstract

Ultra visokotlačno tekočinsko kromatografijo so v laboratorijih razvili v poznih 90 letih prejšnjega stoletja, s ciljem doseganja hitrih analiz visoke ločljivosti pretežno na kapilarnih kolonah napolnjenih z 1–1.7 mikronskimi delci. UHPLC se pogosto uporablja v analizni kemiji za ločevanje, identifikacijo in kvantitativno analizo spojin. Zagotavlja nam veliko hitrost, visoko ločljivost ter občutljivost in kratek čas izvajanja. Za to so potrebni zelo visoki tlaki in sicer do 1500 barov, ter kolone polnjene z delci manjšimi od 2 mikronov. Te lahko razdelimo v dve skupini, na hibridne ter delce na osnovi silike. Prednosti UHPLC sistemov so tudi manjša uporaba organskih topil ter zmanjšanje nastalih odpadkov. Med seboj se UHPLC sistemi razlikujejo po največjem doseženem tlaku, različnih možnostih pretoka, mrtvem volumnu in številnih drugih lastnostih. Dandanes se UHPLC kolone uporabljajo pri okoljskih, živilskih ter kemijskih analizah, kjer je potrebno analizirati veliko število vzorcev v čim krajšem času. Prav tako se zaradi hitrega ločevanja in zmanjšanih stroškov široko uporablja v farmaciji med odkrivanjem in razvojem novih zdravil.

Secondary abstract: Ultra-high-pressure liquid chromatography was developed in laboratories in the late 1990s, with the aim of achieving rapid high-resolution analysis predominantly on capillary columns filled with 1–1.7 micron particles. UHPLC is widely used in analytical chemistry for the separation, identification and quantitative analysis of compounds. It provides us with high speed, high resolution, sensitivity and short lead time. This requires very high pressures of up to 1500 bar and columns filled with particles smaller than 2 microns. We can divide them in two groups, into hybrid and silica-based particles. The advantages of UHPLC systems are the use of less organic solvents and a reduction in generated waste. UHPLC systems differ from each other in maximum pressure drop, different flow rates, dead volume and many other properties. Nowadays, UHPLC columns are used in environmental, food and chemical analyzes, where it is necessary to analyze a large number of samples in the shortest possible time. It is also widely used in pharmaceutical companies during the discovery and development of new drugs due to its rapid separation and reduced costs.
