Abstract
Živimo v času, kjer se vse pogosteje zavedamo, da hrana ni samo zagotavljanje človeških osnovnih potreb, ampak ima velik vpliv tudi na naše zdravje in posledično počutje. Živila ocenjujemo glede na njihov biološki potencial in sposobnost zmanjšanja tveganja za razvoj nekaterih bolezni, med njimi tudi raka.
V magistrskem delu smo pridobivali ekstrakte konoplje z različnimi postopki ekstrahiranja. Uporabili smo konvencionalne metode, kot so ekstrakcija po Soxhletu, hladna maceracija in ultrazvočna ekstrakcija. Pri superktritičnih ekstrakcijah s CO2 smo uporabili različne tlake in temperature. Kot topilo smo uporabili 100% etanol. Preverili smo vpliv postopka ekstrakcije na izkoristke reakcij in določili vsebnosti totalnih fenolov, proantocianidinov, antioksidantov in flavonoidov. Pridobljene ekstrakte smo nanesli tudi na človeške melanomske celice WM-266-4 in preverjali vpliv teh ekstraktov na zmanjšanje metabolne aktivnosti rakastih celic.
Ugotovili smo, da smo največji izkoristek dosegli pri ekstrakciji s Soxhletom in ultrazvočni ekstrakciji, medtem ko smo pri suprekritičnih ekstrakcijah s CO2 dosegli manjše izkoristke. Največjo vrednost totalnih fenolov, flavonoidov in antioksidantov smo dosegli pri ultrazvočni ekstrakciji, medtem ko smo največjo vsebnost proantocianidinov zaznali pri ekstrakciji s Soxhletom. Pri superkritičnih ekstrakcijah s CO2 je bila vsebnost proantocianidinov, totalnih fenolov in antioksidativna učinkovitost višja pri višjem tlaku in temperaturi.
Na koncu smo ekstrakta pridobljena z ultrazvočno in superkritično ekstrakcijo nanesli še na človeške melanomske celice WM-266-4. Ugotovili smo, da so vsi ekstrakti zavirali metabolno aktivnost rakastih celic, saj se je zaustavila delitev celic. Za najboljšega med ekstrakti pa se je v obeh primerih izkazal ekstrakt s koncentracijo 1 mg/mL. Vsem tem ekstraktom smo določili tudi visoko vrednost totalnih fenolov, antioksidantov, proantocianidinov ter totalnih flavonoidov, zato lahko predvidevamo, da so te vrednosti povezane z zaustavitvijo rasti rakastih celic.

We live in an era where we are increasingly conscious of the fact that food isn't just the means to sustain human basic needs, but it influences our physical health and consequently, our mental state, too. Foodstuffs are rated based on their biological potential and the ability to decrease the risk of developing certain diseases, including cancer.
In the master’s thesis, we presented the acquiring of hemp extracts by different processes of extraction. We used conventional methods, such as a Soxhlet extractor, cold maceration and ultrasound extraction. With supercritical extractions with carbon dioxide, we used different pressures and temperatures. As a solvent we used 100 per cent ethanol. We examined the effect of the extraction process on reaction yields and determined the content of total phenols, proanthocyanidins, antioxidants and flavonoids. We laid the acquired extracts on human melanoma cells WM-266-4 and tested the influence of extracts on the reduction of metabolic activity of cancer cells.
We established the biggest yield was achieved with the Soxhlet extractor and the ultrasound extraction, while we achieved smaller yields with supercritical C02 extractions. The highest content of total phenols, flavonoids and antioxidants was reached with the ultrasound extractions, while the highest content of proanthocyanidins was achieved with the Soxhlet extractor. With supercritical C02 extractions, the content of proanthocyanidins, total phenols and antioxidants efficiency was higher when the pressure and temperature were higher.
In the conclusion, we laid the extracts gained through the ultrasound and the supercritical extractions on the human melanoma cells WM-226-4. We determined that all extracts decelerated the metabolic activity of the cancer cells because the cell division ceased. The best among extracts in both cases was the extract with a concentration of 1 mg/mL. All of these extracts were also determined with a high total phenol, proanthocyanidins, antioxidants and total flavonoids content. That is why we can assume these contents are associated with the cease in the growth of the cancer cells. |
