Abstract

Kinurenska kislina je biološko pomembna organska spojina, ki jo med drugim najdemo tudi v medu. Na podlagi pregledane literature sem razvila metodo za določanje kinurenske kisline v medu s tekočinsko kromatografijo z UV in fluorescenčno detekcijo. Kot topilo za vzorce sem izbrala 0,1% vodno raztopino amonijaka, kot mobilno fazo pri kromatografiji pa mešanico acetonitrila in vodne raztopine mravljične kisline. Valovne dolžine svetlobe za detekcijo kinurenske kisline sem določila na podlagi posnetih absorpcijskih in emisijskih spektrov: valovna dolžina absorpcije in vzbujanja pri 240 nm, valovna dolžina emisije pri 410 nm. Analizno metodo sem validirala s določitvijo linearnosti, ponovljivosti, LOD in LOQ. Z metodo umeritvene krivulje sem določila koncentracijo kinurenske kisline v 12 vzorcih medu proizvajalca Medex. Najvišja koncentracija kinurenske kisline je bila v kostanjevem medu, najmanjša pa v akacijevem medu.

Secondary abstract: Kynurenic acid is a biologically important organic compound that can be found in honey. Based on the reviewed literature, a method was developed for determining kynurenic acid content in honey by liquid chromatography with UV and fluorescence detection. 0.1% solution of ammonia in water was selected as the solvent for the samples, and a mixture of acetonitrile and water solution of formic acid as the mobile phase for chromatography. Wavelengths of light for the detection of kynurenic acid were determined on the basis of recorded absorption and emission spectra: absorption and excitation wavelength at 240 nm and emission wavelength at 410 nm. The analytical method was validated by determining linearity, repeatability, limit of detection, and limit of quantitation. Using the calibration curve method, content of kynurenic acid was analysed in 12 samples of Medex honey. The highest concentration was found in chestnut honey and the lowest in acacia honey.
