Abstract

Nikotinamid mononukleotid (NMN), je molekula, ki zaradi svojih farmakoloških učinkov v zadnjem času vzbuja veliko zanimanja na farmacevtskem področju. Ker blagodejno vpliva na potek staranja in z njim povezane bolezni, si številni proizvajalci prizadevajo, da tak prehranski dodatek tudi sami postavijo na trg. Če želijo NMN proizvajati, ga je potrebno znati tudi določiti in meriti njegovo vsebnost, kar je tema moje diplomske naloge. Želela sem raziskati možnosti določevanja tovrstne spojine z uporabo HPLC tehnike, pri čemer sem se osredotočila na iskanje optimalnih pogojev kromatografske analize. Preizkušala sem različni kromatografski koloni, mobilne faze z različnimi pH vrednostmi, različne temperature kolone in gradiente. Na podlagi dobljenih razultatov sem določila optimalne pogoje separacije in metodo okarakterizirala s parametroma linearnosti in ponovljivosti.

Secondary abstract: Nicotinamide mononucleotide (NMN) is a molecule that, due to its pharmacological effects, has recently gained a large interest in the pharmaceutical field. As it has a beneficial effect on aging and related diseases, many manufacturers strive to place such a dietary supplement on the market themselves. However, if they want to produce NMN, it is necessary to be able to determine and measure its content, which is the topic of my thesis. I wanted to explore the possibilities of determining this type of compound using the HPLC technique, focusing on finding the optimal conditions for chromatographic analysis. I tested different chromatographic columns, mobile phases with different pH values, different column temperatures and gradients. Based on the obtained results, I determined the optimal separation conditions and characterized the method with the parameters of linearity and repeatability.
