Abstract

V okviru magistrske naloge smo v prvi fazi raziskav sintetizirali globoko evtektična topila, ki predstavljajo zeleno alternativo konvencionalnim organskim topilom. Topila smo sintetizirali na osnovi holin klorida, kot akceptorja vodikove vezi; propan-1,2-diol, ocetno kislino, mlečno kislino, fruktozo in butan-1,4-diol pa smo izmenično uporabili kot donorje vodikove vezi. Učinkovitost ekstrakcij z globoko evtektičnimi topili smo primerjali z učinkovitostjo ekstrakcij s konvencionalnimi organskimi topili, kot sta metanol in etanol ter deionizirano vodo. Bioaktivne spojine iz uprašenih čebulnih olupkov smo ekstrahirali po metodi ultrazvočne ekstrakcije v kombinaciji s centrifugiranjem in jih identificirali z analizo na tekočinski kromatografiji visoke ločljivosti z UV detektorjem in tekočinski kromatografiji z masnim spektrometrom. Bioaktivne komponente smo kvantitativno ovrednotili z zasnovo umeritvenih krivulj, ki smo jih pripravili iz osnovnih standardnih raztopin. Pri proučevanju bioaktivnih učinkovin iz čebulnih olupkov smo se omejili na določanje protokatehuične kisline, kvercetina, kvercetin-3,4'-diglukozida in kvercetin-3-β-D-glukozida. Dobljenim ekstraktom smo po večih metodah (ABTS, FRAP, DPPH, TPC) določili antioksidativno učinkovitost. Rezultate obeh analiz smo poenotili in ugotovili, da smo s 70 % metanolom in globoko evtektičnimi topili, kjer smo kot donor vodikove vezi uporabili ocetno kislino, mlečno kislino in butan-1,4-diol, ekstrahirali največ spojin z visokim antioksidativnim učinkom.

Secondary title: Deep eutectic solvents for green and sustainable extraction of bioactive compounds from onion (Allium cepa L.) peel
Secondary abstract: As a part of the master's thesis, we synthesized deep eutectic solvents in the first phase of the research, which are an environmentally friendly alternative to conventional organic solvents. The solvents were synthesized based on choline chloride as a hydrogen bond acceptor; propane-1,2-diol, acetic acid, lactic acid, fructose, and butane-1,4-diol were separately used as hydrogen bond donors. The efficiency of extractions with deep eutectic solvents was compared with the efficiency of extractions with conventional organic solvents such as methanol and ethanol, and deionized water. Bioactive compounds from powdered onion peels were extracted by ultrasonic extraction combined with centrifugation and identified by high-performance liquid chromatography with UV detector and liquid chromatography with a mass spectrometer. Bioactive components were quantitatively evaluated by generating calibration curves prepared from standard solutions. In the study of bioactive compounds from onion peels, our research was limited to the determination of protocatechuic acid, quercetin, quercetin-3,4'-diglucoside, and quercetin-3-β-D-glucoside. The antioxidant activity of the extracts obtained was determined by different methods (ABTS, FRAP, DPPH, TPC). Based on the results it was concluded that 70 % methanol and deep eutectic solvents using acetic acid, lactic acid, and butane-1,4-diol as hydrogen bond donors extracted compounds with the highest antioxidant activity.
