Abstract

Cilj diplomskega dela je bila sinteza peptidov z uporabo dveh različnih metod. Oba načina sta temeljila na principu SPPS (ang. Solid-phase peptide synthesis) in uporabi Fmoc metode. Pri prvem načinu smo za sintezo uporabili inštrument na osnovi pretočne SPPS, ki je omogočal hitro kemijsko sintezo peptidov. Drugi način sinteze je potekal z uporabo zbiralne vakuumske komore, za izvedbo sinteze so bili potrebni daljši časi. Uspešno smo sintetizirali tri peptide, ki so vsebovali 17 in 26 aminokislin, od katerih sta bila dva enaka, toda sintetizirana po drugačni strategiji, enkrat s pretočno SPPS, drugič ročno z uporabo zbiralne vakuumske komore. S pretočno SPPS smo peptide sintetizirali v bistveno krajšem času, kot z uporabo zbiralne vakuumske komore, vendar so bili ti bistveno manj čisti.

Secondary abstract: The aim of this diploma thesis was the synthesis of peptides using two different methods. Both methods were based on the SPPS (Solid-phase peptide synthesis) principle and used the Fmoc protected amino acids. In the first method, an instrument based on flow SPPS was used for the synthesis, which enables rapid chemical synthesis of peptides. Another method of synthesis was a much slower standard SPPS method using a collection vacuum chamber. We successfully synthesized three peptides containing 17 and 26 amino acids, two of which were identical, but synthesized using a different strategy, once with flow SPPS, the second time manually using a collection vacuum chamber. With flow SPPS, peptides were synthesized in a significantly shorter time, than using a collection vacuum chamber, but these were significantly less clean.
