Abstract
Melanom je eno izmed najtežje ozdravljivih rakavih obolenj, saj so človeške melanomske celice znane kot ene najbolj agresivnih rakastih celic. Znanstveniki po celem svetu si zato prizadevajo, da bi razvili zdravilo, ki bi lahko ozdravilo to agresivno obliko raka. Dosedanje študije so pokazale, da imajo naravne spojine, med njimi sekundarni metaboliti, najdeni v naravnih materialih oz. proizvodih, tudi v čebeljem cvetnem prahu, citotoksični, antiproliferativni ter kemopreventivni učinek na različne rakave celične linije in so vključene v veliko klinično odobrenih zdravil proti raku.
S pomočjo različnih ekstrakcijskih metod (ultrazvočna ekstrakcija, ekstrakcija po Soxhletu, superkritična ekstrakcija, vroča maceracija in hladna maceracija) smo pridobili ekstrakte cvetnega prahu s 70-odstotnim etanolom kot topilom in jim določili vsebnost totalnih fenolov, proantocianidinov in antioksidantov. Z računalniškim programom Design Expert smo optimirali pogoje ultrazvočne ekstrakcije in raztopine različnih koncentracij teh ekstraktov nanesli na melanomske celice. Rezultati so pokazali, da so vsi ekstrakti vplivali na zaviranje rasti rakavih celic, saj so zaustavili njihovo delitev, vendar opazimo signifikantno zmanjševanje metabolne aktivnosti rakastih celic pri koncentracijah med 0,05 mg/mL in 0,5 mg/mL.

Influence of optimised pollen extract on metabolic activity of WM-266-4 melanoma cells |

Melanoma is one of the hardest types of cancer(s) to treat because melanoma cells are known as some of the most aggressive cancer cells in existence. That is why scientists all over the world are striving to develop a cure for this particularly ugly and aggressive cancer. Studies up to now have shown that natural compounds, which include secondary metabolites found in natural materials/products (even in bee pollen), have cytotoxic, antiproliferative, and chemopreventive effects on various cancer cell lines and are consequently incorporated into many clinically approved oncology (anti-cancer) drugs. With the help of various different extraction methods (ultrasonic extraction, Soxhlet extraction, supercritical extraction and lastly hot and cold maceration) we have been able to acquire, with 70% ethanol serving as a solvent, pollen extractions- and assign to them total phenolic, proanthocyanidin and antioxidant content. With the help of the ‘Design Expert’ computer program we have optimized the conditions of ultrasonic extraction and have applied extraction solutions of different concentrations to melanoma cells. The results show that all extractions had an effect on the growth of cancer cells; they stopped their division, however we can observe a significant reducement in cancer metabolism, when we approach concentrations between 0,05 and 0,5 mg/mL. |
