Abstract

V diplomskem delu obravnavamo energijska stanja v atomih in molekulah, lastnosti fluorescence, fosforescence in sipanja kot prehode med molekulskimi energijskimi stanji. Opišemo tudi Beer-Lambertov zakon za vse tri pojave. Meritve opravimo s spektrofotometrom MUT Tristan Light in s preoblikovano transmisijsko komoro. Oprema je bila nabavljena v okviru Norveškega finančnega mehanizma. Z meritvami pokažemo Stokesov premik za fluorescenco, spektralni maksimum za fosforescenco in odvisnost intenzitete svetlobe od valovne dolžine. Z meritvami tudi pokažemo, da je prek Beer-Lambertovega zakona mogoče izračunati neznano koncentracijo raztopine tudi iz gostot fluoresciranega oziroma sipanega svetlobnega toka vzorcev. Del diplomskega dela je namenjen zgodovini raziskovanj pojavov in njihovim aplikacijam v moderni znanosti.

Secondary abstract: In the thesis we study and describe the energy levels of atoms and molecules, possible transitions between them, properties of fluorescence, phosphorescence and scattering from the point of view of transitions between molecular energy states. We also present the original form of Beer-Lambert's law as well as its modifications and applications in all three studied phenomena. The measurements were performed with the spectrometer MUT Tristan Light and the transformed transmission chamber. The equipment was acquired within the project financed from the Norway Grants. With the spectrophotometer we can measure the Stoke's fluorescence shift, phosphorescent spectral maximum and the dependence of the intensity of the scattered light with respect to their wavelengths. From the measured spectra of fluoresced or scattered light we were able to determine the concentrations of either fluorescent dyes or small particles in the liquid samples with the use of the modified Beer-Lambert's law. A part of the thesis is dedicated also to the presentation of a historical overview of these phenomena as well as of their applications in modern science.
