Abstract

Hidrolati so stranski produkti pri pridelavi eteričnih olj. Proučevali smo vpliv hidrolatov damaščanske vrtnice, sivke in laškega smilja v dodanih celičnih kulturah keratinocitov NHEK in melanomskih celic WM-266-4 na obliko celic in njihovo metabolno aktivnost. Pripravili smo gojišča za celični kulturi keratinocitov NHEK in melanomskih celic WM-266-4. Celice smo izpostavili raztopinam hidrolatov. S pomočjo svetlobne mikroskopije smo izvedli morfološko analizo, s testom WST-8 analizo metabolne aktivnosti. Z morfološko analizo smo ugotovili spremenjeno obliko keratinocitov NHEK v celičnih kulturah, v katere smo dodali hidrolat sivke in laškega smilja. Metabolna aktivnost keratinocitov NHEK se je znižala pri hidrolatu sivke in laškega smilja. Hidrolat damaščanske vrtnice ni imel vpliva na obliko in metabolno aktivnost keratinocitov NHEK, prav tako ni vplival na melanomske celice WM-266-4. Hidrolata sivke in laškega smilja sta vplivala na obliko melanomskih celic WM-266-4. Na obliko celic je imel hidrolat sivke večji vpliv. Hidrolata sivke in laškega smilja sta povzročila inhibicijo metabolne aktivnosti melanomskih celic WM-266-4.

Secondary abstract: Hydrolates are by-products of essential oil production. The effect of hydrolates of Damascene rose, lavender, and immortelle in added cell cultures of NHEK keratinocytes and WM-266-4 melanoma cells on cell shape and metabolic activity was investigated. Cell culture media for NHEK keratinocytes and WM-266-4 melanoma cells were prepared. The cells were exposed to hydrolat solutions. Morphological analysis was performed by light microscopy and metabolic activity was analysed by WST-8 assay. The morphological analysis revealed an altered shape of NHEK keratinocytes in cell cultures supplemented with lavender and immortelle hydrolates. The metabolic activity of NHEK keratinocytes decreased with lavender and immortelle hydrolates. Damascene rose hydrolat did not affect the shape and metabolic activity of NHEK keratinocytes, nor did it affect WM-266-4 melanoma cells. Lavender and immortelle hydrolates had no effect on the shape of WM-266-4 melanoma cells. Lavender hydrolates had a greater effect on cell shape. Lavender and immortelle hydrolates caused inhibition of the metabolic activity of WM-266-4 melanoma cells.
