Secondary abstract: Aggregatibacter actinomycetemcomitans is a bacterium that causes aggressive and chronic parodontitis. The main effect of this periodontal disease is a collapse of periodontal tissues in a very short period of time. This is caused by virulent factors that the bacteria excrete in periodontal pockets. Most known factors are leukotoxin and toxin CDT (Cytolethal distending toxin). In my thesis I have focused on the bacteria A. actinomycetecomitans and associated it with parodontitis disease. Most of my work consisted of the preparation of plasmid construct to examine the promoter activity for the biosynthesis of leukotoxin and toxin CDT in bacteria. By engineering the recombinant plasmid that harbours the promoter region of the toxin CDT or of the leukotoxin fused to the β-galactosidase reporter gene we can recognize the transcriptional factors that are important for regulation of the gene expression and therefore determine the environmental signals needed for the induction of either toxin.

Abstract (ePrints): Aggregatibacter actinomycetemcomitans je bakterija, ki povzroča agresivni in kronični parodontitis. Glavna posledica te parodontalne bolezni je propad obzobnih tkiv v zelo kratkem času. Bolezensko stanje se pojavi zaradi virulenčnih faktorjev, ki jih bakterija izloča v parodontalne žepe, med katerimi sta najbolj znana toksina CDT (ang. Cytolethal distending toxin) in levkotoksin. V diplomskem delu sem se posvetila bakteriji A. actimomycetemcomitans ter jo povezala z boleznijo parodontitis. Osredotočila sem se na pripravo plazmidnih konstruktov za preučitev promotorske aktivnosti za biosintezo levkotoksina in toksina CDT v bakteriji. S plazmidnim konstruktom, ki vsebuje promotorsko regijo toksina CDT ali levkotoksina spojenega s poročevalskim genom za β-galaktozidazo, lahko določimo dejavnike, ki vplivajo na izražanje genov teh toksinov. Posledično lahko določimo signale okolja, ki so pomembni za sintezo teh toksinov.
