Secondary title: NLP proteins activity on tobacco leaves
Secondary abstract: Some bacteria, fungi and oomycetes can produce NLP proteins (Nep1-like proteins), which induce the release of ethylene, cause tissue necrosis and trigger a hypersensitive response. Since NLP proteins only affect dicotyledons and are also known to cause tissue necrosis on tobacco leaves, tobacco (Nicotiana tabaccum) has been used as a test plant. Tissue necrosis, rotting and blight are only some of the numerous symptoms that appear as a consequence of dicotyledonous infection with a pathogenic organism. Two recombinant proteins PaNIE and NPP, obtained by means of an expression system in the yeast Pichia pastoris, have been used in this research. The aim is to show that both proteins will cause necrosis on tobacco leaves in nM concentrations. PaNIE protein is produced by pathogenic oomycete Pythium aphanidermatum, while NPP protein is produced by pathogenic oomycete Phytophthora parasitica. We wanted to test the activity of some organic molecules on tobacco leaves both in the presence and in the absence of the protein. Since the oomycetes that produce the mentioned proteins cause severe material damage on crops, we decided to explore the products used to exterminate pathogenic organisms. The research has been limited to Slovene market.
