Abstract

Namen diplomske naloge je bil uspešno imobilizirati encim na biooglje, nosilec, ki je bil pripravljen s hidrotermično karbonizacijo odpadne biomase. S spreminjanjem koncentracije encima in z različno izbiro mrežnega povezovalca smo želeli izvesti imobilizacijo encima na biooglje, ki nam da najvišjo preostalo aktivnost encima. Zanimalo nas je, kaj se z aktivnostjo imobiliziranega encima dogaja pri izpostavitvi različnim dejavnikom kot sta temperatura in čas skladiščenja. Rezultati kažejo, da najvišjo preostalo aktivnost obdrži imobiliziran encim s koncentracijo 0,05 g/mL ter ob uporabi mrežnega povezovalca gluteraldehida, koncentracije 2 %. Čas imobilizacije encima je bil 24 ur. Na stresalniku smo uporabili hitrost stresanja 400 rpm. Pri izpostavitvi imobiliziranega encima določeni temperaturi smo ugotovili, da dalj časa je izpostavljen višji temperaturi, bolj bo njegova aktivnost padala. Pri različnem času skladiščenja pa smo prišli do zaključka, da aktivnost encima s časom skladiščenja prav tako pada.

Secondary abstract: The purpose of the thesis was a successful immobilization of enzymes on biochar, a carrier, which was prepared from hydrothermal carbonization processes of biomass. By varying the concentration of the enzyme with a different choice of crosslinking agent, we want to perform immobilization of the enzyme on biochar, which gives the highest residual activity of the enzyme. We wanted to know, what is happening with the activity of the immobilized enzyme, when exposed to different conditions such as temperature and time of storage. The results show that the maximum residual activity of the immobilized enzyme was detected at enzyme concentration of 0.05 g/mL, using the crosslinking agent gluteraldehide, concentration of 2 %. The immobilization time of the enzyme was 24 hours. On a shaker, we used the agitation rate of 400 rpm. During the exposal of immobilized enzyme to a certain temperature, we found out that with the prolongation of exposition time of immobilized enzyme to the higher temperature, the activity of enzyme decreased. It was also detected that with the increase of storage time, the activity of immobilized enzyme decreased.
