Abstract

V magistrskem delu smo proučevali vpliv tlaka in časa inkubacije celične suspenzije črne kvasovke Phaeotheca triangularis v superkritičnem ogljikovem dioksidu. Celično suspenzijo smo inkubirali pri temperaturi 35 °C in pri različnih tlakih in časih inkubacije v SC CO2 in sicer pri 100 bar, 200 bar in 300 bar, ter časih izpostavitve 0,5 ure, 1 uro, 2 uri in 3 ure. Zanimalo nas je kako tlak in čas izpostavitve celične suspenzije P. triangularis v SC CO2 vplivata na aktivnost nekaterih encimov: α-amilaze, β-glukozidaze, celulaze in proteaze, na razmerje med nukleinskimi kislinami in proteini ter koncentracijo proteinov. Z razmazi celične suspenzije pred in po inkubaciji v SC CO2 na gojišča pa smo določevali še preživelost celične kulture P. triangularis. Ugotovili smo, da z naraščanjem tlaka in časa inkubacije celične suspenzije v SC CO2, preživelost celic P. triangularis upada. Tudi preostala aktivnost encimov se je spreminjala. Preostali aktivnosti celulaze in α-amilaze z naraščanjem časa izpostavitve celične suspenzije SC CO2 naraščata, medtem ko preostali aktivnosti β-glukozidaze in proteaze upadata. Preostale aktivnosti vseh preučevanih encimov z naraščanjem tlaka izpostavitve upadajo. Koncentracija proteinov v celični suspenziji P. triangularis z naraščanjem časa in tlaka narašča. Z določevanjem razmerja med nukleinskimi kislinami in proteini pa smo ugotovili, da so v celični suspenziji prisotni še proteini, fenoli ali druge snovi.

Secondary abstract: The influence of pressure and incubating time on cell suspension of black fungi Phaeotheca triangularis in supercritical carbon dioxide was studied. Cell suspension was incubated at temperature 35 °C and by different pressures (100 bar, 200 bar and 300 bar) for 0,5 hour, 1 hour, 2 hours and 3 hours. The impact of pressure and incubation time on activity of some enzymes: α-amylase, β-glucosidase, cellulase and protease, in addition, on the ratio between nucleic acids and proteins and also on the concentration of proteins was investigated. With the smear of the cell suspension before and after incubation in SC CO2 on the agar, the viability of the cell culture P. triangularis was determined. With an increase in pressure and incubation time of cell suspension in SC CO2 viability of P. triangularis decreased. The residual enzyme activity also varied. Residual activities of cellulase and α-amylase were increased by increase in incubation of cell suspension in SC CO2, while residual activities of β-glucosidase and protease were decreased. With increasing of pressure the residual activities of all studied enzymes were decreased. The protein concentration in the cell suspension of P. triangularis increased by increase in incubation time and pressure. By determining the ratio between nucleic acids and proteins, we have found that in the cell proteins, phenols or other substances are presented.
