Abstract
Namen diplomskega dela je bil razvoj in validacija analizne metode za identifikacijo in določevanje alkoholov v različnih vrstah vina.
Alkoholi nastajajo v moštu med procesom fermentacije oziroma alkoholnega vrenja. Pri določevanju alkoholov smo se osredotočili predvsem na dve vrsti alkoholov in sicer na metanol in etanol. Določevanje vsebnosti metanola, ki sicer ne nastaja pri fermentaciji, pač pa je sestavni del pektinskih snovi, kjer je zaestren na polimer galakturonske kisline, je s stališča raziskovalca zanimivo, saj je metanol strupen že v majhnih količinah, njegova vsebnost pa ni označena na steklenici. Določene vsebnosti etanola pa smo primerjali z deklariranimi vsebnostmi, podanimi na etiketah steklenic in tako preverili točnost podatkov.
Kot metodo za določevaje alkoholov v vinih smo uporabili plinsko kromatografijo v povezavi s plamensko – ionizacijskim detektorjem. Pri razvoju analizne metode smo pripravili raztopine standarda etanola in metanola, ter raztopine vzorcev treh različnih vin. Raztopino standarda etanola smo z redčenjem pripravili dvanajst odstotno, saj je vsebnost etanola v povprečnem vinu okoli deset odstotkov, medtem ko smo raztopino standarda metanola pripravili na precej nižji koncentraciji (okrog 0,1 odstotno), saj metanola vina skoraj ne vsebujejo.
Analizno metodo smo nato validirali, saj se le validirana analizna metoda lahko uporablja za namen raznih analiznih določitev.Validacija je proces pri katerem z različnimi eksperimentalnimi testi pokažemo, da neka metoda ustreza vsem predpisanim zahtevam za njeno uporabo.
Validacijska študija je pokazala, da je metoda natančna, točna, selektivna, linearna, robustna in rigidna in tako primerna za uporabo.
Rezultati analize kažejo, da so vsebnosti etanola v testiranih vzorcih vin primerljive podatkom na etiketah steklenic.

The object of my thesis was development and validation of analytical method for identification of alcohols in different types of wines.
Alcohols are produced in cider during the process of fermentation. When determining types of alcohols in wines I have focused on two specific alcohols; methanol and ethanol. The content of methanol, which is not produced by fermentation but is bonded with ester bond on a polymer of galacturonic acid as a part of pectic substances, is important as it is toxic in small amounts but it is not labeled on the bottle. The content of ethanol is labeled on the bottle, but it was not for sure how accurate that percentage is.
Method used for determination of alcohols was gas chromatography. When developing analytical method, standard solutions of ethanol and methanol were prepared together with solutions of three different types of wines. Ethanol standard was 12 % solution since wines usually contain around ten percent of ethanol and methanol standard was 0.1 % solution as wines usually do not contain high amounts of methanol.
Because only validated analytical method can be used for purposes of analysis, method was also validated. Validation is the process where several experiments are conducted in order to prove that method meets all necessary regulations. Validation study proved the method to be precise, exact, selective, linear, robust and rigid and in that matter suitable for analysis of alcohols present in wines.
Results of analysis show that amounts of ethanol in tested wine samples are comparable with data indicated on bottle labels. |
