Abstract
Namen magistrskega dela je bilo ugotoviti, katero topilo – vodna raztopina acetona ali vodna raztopina etanola je boljša izbira za pripravo produkta ekstrakcije, ki je čim bolj primerljiva s komercialnim piknogenolom.
Za slednjega se ve, da je pripravljen iz obmorskega bora, Pinsu Pinaster, ki je sestavljen iz 50 učinkovin, le-te pa delujejo antioksidacijsko. Med pomembnimi fenolnimi spojinami so proantocianidini, flavonoidi, ki pa so prisotni v manjši meri. Piknogenol naj bi učinkoval na krvožilni sistem, zato se ga priporoča diabetikom. Lajšal naj bi tudi težave z vidom in pospeševal regeneracijo manjših površinskih poškodb kože.
Opravili smo raziskavo ekstrakcij, ki so bile izvedene pri različnih temperaturah - vodne raztopine acetona so bile izvedene pri temperaturi 30, 40 ter 50 °C, vodne raztopine etanola pa pri temperaturi 40, 60 in 80 °C.
Antioksidativnost ekstrakta, vsebnost skupnih fenolov, proantocianidinov smo določevali s pekrtofotometričnimi metodami. Vsebnost komponent, kot so katehin, kavna kislina, ferulna kislina in taxifolin, pa je bila določena s tekočinsko kromatografijo.
Ob primerjavi rezultatov ekstraktov s piknogenolom, ki se uspešno trži, so je izkazal ekstrakt, ki je bil pripravljen z vodno raztopino etanola v razmerju etanol / voda: 10 / 90, (v/v) pri temperaturi ekstrakcije 40 °C, kot najbolj primerljiv s komercialnim piknogenolom.

Purpose of the thesis was to find out which solvent (aqueous solution of acetone or aqueous solution of ethanol) is better for preparing a product of extraction which is the most comparable with commercial pycnogenol.
It is widely known that pycnogenol is produced from Pinsu Pinaster, which is composed of 50 substances that work as antioxidants. Among the important phenol chemical compounds are proanthocyanidins and flavonoids, but are existent in smaller amounts. Pycnogenol affects cardio-vascular system and is recommended to diabetics. It also has effects on sight and regeneration of surface skin wounds. We have conducted a research of extractions which were ran under different temperatures; aqueous solutions of acetone were extracted under the temperatures of 30, 40 and 50 °C and aqueous solutions of ethanol were extracted under the temperature of 40, 60 and 80 °C.
Antioxidativity of the extract and content of phenols and proanthocyanidins was verified with spectrophotometrical methods. Contents of components like catechin, caffeic acid, ferulic acid and taxifolin were verified with liquid chromatography.
Comparison between the extracts and pycnogenol showed that the extract prepared with aqueous solution of ethanol in ratio of ethanol and water is 10/90 (v/v) prepared under the temperatue of 40 °C is the most similar to commercial pycnogenol. |
