Povzetek
Clostridioides difficile (v nadaljevanju C. difficile) je anaerobna, po Gramu pozitivna bakterija, ki se zaradi prilagajanja na neugodne okoljske razmere preoblikuje v spore. Spore so zelo odporne in omogočajo dolgoročno preživetje v različnih okoljskih nišah, kot so zemlja, kompost in voda. Genomske študije so pokazale, da se populacija C. difficile diferencira in jo je mogoče razdeliti v več kladov (filogenetskih skupin): klade od 1 do 5 in divergentne klade od C-I do C-V. Sevi iz divergentnih kladov se genotipsko zelo razlikujejo od sevov iz kladov 1–5 in jih pogosteje najdemo v okolju, čeprav so bili opisani primeri okužb s sevi iz divergentnih kladov. Sevi iz kladov 1–5 so prisotni pri ljudeh, živalih in v različnih naravnih okoljih. Veliko raziskav o aktivnosti C. difficile je narejenih na obeh gostiteljih, človeku in živali. V okviru magistrskega dela smo z gojitvenimi metodami skušali preveriti prisotnost aktivnih oblik C. difficile v okolju izven gostitelja. Gojenje smo uporabili za preverjanje rasti C. difficile v nadzorovanih pogojih, tj. v vzorcih komposta. V magistrsko delo smo vključili tri kompostnike z različnih lokacij. Prisotnost C. difficile smo potrdili v enem. Ta kompost smo uporabili za pripravo mikrokozmov in spremljanje rasti C. difficile. Dobljeni rezultati potrjujejo postopno rast C. difficile v mikrokozmosih, s čimer smo prvi pokazali, da se lahko bakterija C. difficile razmnožuje v kompostu.

Clostridioides difficile (C. difficile) is an anaerobic, Gram-positive bacterium that transforms into spores to adapt to adverse environmental conditions. The spores are highly resistant and allow long-term survival in different environmental niches such as soil, compost and water. Genomic studies have shown that the C. difficile population is differentiating and can be divided into several clades (phylogenetic groups): clades from 1 to 5 and divergent clades from C-I to C-V. Strains from divergent clades are genotypically very different from strains from clades 1–5 and are more frequently found in the environment, although cases of infection with strains from divergent clades have been described. Strains from clades 1–5 are present in humans, animals and in a variety of natural environments. Much research on the activity of C. difficile has been carried out in both human and animal hosts. In the framework of this MSc thesis, we wanted to test the presence of active C. difficile in a non-host environment, using cultivation methods. Cultivation was used to verify the growth of C. difficile under controlled conditions, i.e. in compost samples. In the master thesis we included three composters from different locations. The presence of C. difficile was confirmed in one. This compost was used to prepare microcosms and monitor the growth of C. difficile. The results obtained confirm the gradual growth of C. difficile in the microcosms over time, which is the first demonstration that C. difficile can proliferate in compost. |
