Povzetek
V magistrskem delu je predstavljena sinteza supermagnetnih nanodelcev Ni67,5Cu32,5 v matrici silike s pomočjo sol-gel metode. Nanodelcem s pomočjo silikatne prevleke zagotovimo netoksičnost in biokompatibilnost hkrati pa nam omogoča mesto, kjer vgradimo zdravilno učinkovino.
S pomočjo nanodelcev želimo doseči dvojno terapijo, in sicer preko kombinacije magnetne hipertermije, ki jo omogočajo delci sami, hkrati pa bi lahko s pomočjo magnetnega polja delce tudi usmerjali na tarčno mesto, kjer bi lahko prišlo do nadzorovanega sproščanja vgrajene zdravilne učinkovine (ZU). V ta namen smo v sintetizirane nanodelce vgradili modelne, paracetamol in protitumorsko učinkovino, paracetamol. Iz rezultatov in vitro študije, ki je temeljila na sproščanju ZU s pomočjo UV/VIS spektrometra lahko potrdimo uspešno vgradnjo ZU. V testiranju biokompatibilnosti s humanimi kožnimi celicami pa lahko podpremo literaturo, ki navaja da nanodelci s protitomursko učinkovino uspešno »pobijajo« celice bazalno-celičnega karcinoma.
Sintetizirane magnetne nanodelce smo karakterizirali s pomočjo rentgenske praškovne difrakcije (XRD), termogravimetrične analize (TGA), transimisijske elektronske mikroskopije (TEM), Fourier transformirane infrardeče spektroskopije (FTIR) ter določali profil sproščanja vgrajenih zdravilnih učinkovin.

This master's degree presents a synthesis of the superparamagnetic particles Ni67,5Cu32,5 in a matrix of silics with the help of the sol-gel method. The nanoparticles are provided nontoxicity and biocompatibility via a silica coating. At the same time, there arises space, where you can build in an active substance.
The aim of this degree is to achieve a combination therapy. Firstly, it can be achieved via a combination of the magnetic hyperthermia, enabled by its own particles. Secondly, the particles can be directed at a target with the help of a magnetic field at the same time. At this targeted place, the controlled release of the in-built active substance could happen. For this reason, fluorouracil has been built into the synthesised nanoparticles. Fluorouracil is, namely, paracetamol and a medication treating cancer.
The results of the in vitro study, which has been based on a release of the active substance with the help of the UV/VIS spectrophotometre, confirm the active substance being built in successfully.
When testing biocompatibility of the human skin cells, this degree agrees with the literature which cites that nanoparticles with antitumour substance successfully kill the cells of the basal-cell carcinoma.
There have been many procedures used when synthesised magnetic nanoparticles have been characterized and when the profile of the release of the in-built active substances has been defined. Namely, they are the x-ray powder diffraction (XRPD), the thermogravimetric analysis (TGA), the Transmission Electron Microscopy (TEM) and the Fourier - transform infrared spectroscopy (FTIR). |
