Povzetek
Fiziologija in patofiziologija eksokrinega dela trebušne slinavke sta v tesni povezavi s spremembami znotrajcelične koncentracije kalcijevih ionov. Večina našega znanja o kalcijevih signalnih poteh v acinarnih celicah temelji na poskusih in vitro, kjer celice encimsko izoliramo, kar spremeni njihovo strukturo, fiziologijo in omeji naše razumevanje. Zaradi teh omejitev smo uvedli metodo tkivne rezine trebušne slinavke kot eksperimentalni model in situ. Ocenili smo sposobnost za preživetje in morfološke značilnosti acinarnih celic z uporabo testa preživetja celic (LIVE/DEAD assay), transmisijske elektronske mikroskopije in imunofluorescenčnih metod. V glavnem delu raziskave smo opredelili funkcionalni odziv acinarnih celic na stimulacijo z acetilholinom in ga primerjali z odzivi na cerulein. Spremembo znotrajcelične koncentracije kalcijevih ionov smo merili s konfokalno mikroskopijo. Pokazali smo, da so različni parametri kalcijevih oscilacij koncentracijsko odvisni in da je aktivnost precej dobro usklajena znotraj acinusov, ne pa med acinusi. Metoda tkivne rezine trebušne slinavke tako v naših očeh predstavlja pomemben nov in zanesljiv eksperimentalni model za oceno normalne in patološke morfologije in fiziologije acinarnih celic.

The physiology and pathophysiology of the exocrine pancreas are closely related to changes in intracellular calcium concentration. Most of our knowledge of calcium signaling pathways in acinar cells is based on in vitro experiments where cells or acini are enzymatically isolated from their environment, which can change their structure and physiology and limit our understanding. Due to these limitations, the pancreatic tissue slice technique was introduced as an experimental in situ model. The viability and morphological characteristics of acinar cells within the tissue slice were assessed using the LIVE/DEAD assay, transmission electron microscopy, and immunofluorescence imaging. In the main part of the study, we define the response of acinar cells to acetylcholine stimulation and compare them with the responses to cerulein. Changes in intracellular calcium concentration were detected by confocal microscopy. We have shown that the different parameters of calcium oscillations are concentration-dependent and that the activity is rather well synchronized within acini but not between acini. The pancreatic tissue slice technique represents in our eyes an important, new and reliable experimental model for evaluating the normal and pathological morphology and physiology of acinar cells. |
