Povzetek

V sklopu tega diplomskega dela smo izvajali eksperimente na vrsti tradeskancija (Tradescantia fluminensis) v laboratorijih oddelka za biologijo na Fakulteti za naravoslovje in matematiko v Mariboru. Eksperimentalno delo je potekalo od začetka marca do začetka aprila v letu 2012. Omenjeno vrsto smo izpostavili dvema stresnima dejavnikoma, suši in pomanjkanju svetlobe. V ta namen smo uporabili 4 vzorce za en stresni dejavnik in 4 za drugega ter ob tem uporabili še dva kontrolna vzorca. Skupaj smo tako imeli 10 vzorcev. Meritve, ki smo jih opravljali 30 dni, so zajemale fluorescenco zelenega barvila, ki smo jo merili z merilcem fluorescence klorofila Handy PEA (Hansatech), vsebnost zelenega barvila v listih, izmerjenih s pomočjo merilca vsebnosti klorofila SPAD - 502 (Minolta), odstotek vlage v tleh, pridobljenih z napravo za merjenje vlage HH2 (Delta - T). Merili smo prav tako dolžino poganjka, dolžino zadnjega internodija ter maksimalno širino in dolžino lista, na katerem smo opravljali meritve fluorescence. Tekom eksperimenta smo šteli tudi liste: vse liste na rastlini, zelene zdrave liste, liste s suhim robom in suho konico ter suhe liste. Na začetku izvajanja eksperimentov smo izmerili še svetlobne pogoje, v katerih smo imeli vzorce, ter temperaturo prostora. Bistvo oz. sklep eksperimentalnega dela je, da se je Tradescantia fluminensis odzivala na stresne dejavnike (pomanjkanje vode in svetlobe) ter tako izrazila fiziološke in morfološke spremembe. Naši rezultati so pokazali različne odgovore T. fluminensis na oba stresorja. Sušo tolerira bolje od teme. Prvi pridobljeni odgovor je bila fluorescenca klorofila, nato vrednosti SPAD in na koncu še morfometrične značilnosti. Rastline v suši so bile manjše z manjšimi listi, večjim številom poškodovanih listov in nižjimi vrednostmi SPAD. Rastline brez svetlobe so kazale hitre odgovore upadanja količnika Fv/Fm. Te imajo daljše korenine in liste ter propadejo hitreje kot rastline v suši.

Sekundarni povzetek: Experimental work on Tradescantia fluminensis, included in this thesis, was performed at the Biology department of the Faculty of Natural Sciences and Mathematics from March until April 2012. We were growing plants under stressful conditions: drought and low-light. We used 4 samples (clones) for each stressor and two for control. Measurements of plant performance under stress included chlorophyll fluorescence measurements with portable device Handy PEA (Hansatech) and evaluation of chlorophyll content via green color of the leaf obtained with SPAD - 502 (Minolta). Measurements were recorded daily for 30 days. Other measurements included relative water content in the soil (HH2 device from Delta - T), light conditions - photosynthetically active radiation (PAR; quantum meter Apogee Instruments), and temperature. Additionally, we performed morphometrical measurements by recording the length of the shoot, length of the last shoot segment, as well as maximal leaf length and width. The same leaves were used for fluorescence and morphometrical measurements. Additional observations of visible damage on leaves (dry edge, dry tip, chlorosis, necrosis) were included. Our results show different responses of T. fluminensis to both stressors. Drought is tolerated more than low-light conditions. First response was observed by chlorophyll fluorescence, than SPAD value and at last, morphological characteristics. Plants in drought were smaller with smaller leaves and higher numbers of damaged leaves and the lowest SPAD value. Plants without light showed the fastest response of Fv/Fm decline. They had longer shoots and leaves, however, leaves and plants died sooner than plants in drought.
