Povzetek
Namen magistrskega dela je bil proučiti vpliv različnih načinov predobdelave surove celuloze na učinkovitost encimske hidrolize.
Pri delu smo uporabljali mikrokristalinično celulozo, celulozo iglavcev (iglavci Pacifico), listavcev (listavci Bukocell) in evkaliptusa (evkaliptus Santa Fe). Preizkusili smo tri metode predobdelave. Testirali smo predobdelavo celuloze z ionsko tekočino - BMIMCl, s H2O2 ter avtoklaviranje v raztopini NaOH.
Encimsko hidrolizo smo izvajali v šaržnem reaktorskem sistemu EasyMax 102 (Mettler Toledo). Eksperimente smo izvajali 24 h pri temperaturi 45 C in pri vrtilni hitrosti mešala 200 min-1, uporabili smo med 0,5 in 1 mL encima celulaze in 50 mL acetatnega pufra s pH = 5. Koncentracijo nastale glukoze smo določili s spektrofotometrično metodo in uporabo UV-VIS spektrofotometra (Varian Cary 1-E UV-Visible).
Pri delu smo postopke predobdelave optimirali, saj smo tako zagotovili ustrezne procesne pogoje za večjo presnovo. Največjo presnovo celuloze smo dosegli po predobdelavi z ionskimi tekočinami. Presnova obdelane celuloze se je glede na presnovo neobdelane celuloze povečala iz 10 % na 62 %.

The aim of this work was to investigate the effect of different pretreatment methods of raw cellulose on enzymatic hydrolysis efficiency.
During experiments we used microcrystalline cellulose, cellulose of conifers (pines Pacifico), hardwood (deciduous Bukocell) and eucalyptus (Eucalyptus Santa Fe). We tested three methods of pre-treatment: the pretreatment with the ionic liquids - BMIMCl and H2O2, and autoclaving with the solution of NaOH.
Enzymatic hydrolysis was carried out in a batch reactor system Easymax 102 (Mettler Toledo). Experiments were performed for 24 h at temperature of 45 °C and at rotational speed of the stirrer 200 min-1, using between 0,5 and 1 mL of the enzyme cellulase, and 50 mL of acetate buffer with pH = 5. The concentration of the resulting glucose was determined by a spectrophotometric method, using the UV-VIS spectrophotometer (Varian Cary 1 E UV-Visible).
The pretreatment procedures were optimized, because adequate process conditions for increasing metabolism had to bo ensured. The highest conversion of cellulose was achieved by pretreatment with the ionic liquid. Conversion of pretreated cellulose was increased from 10 to 62% compared to untreated one. |
