Abstract

Rebrinčevolistna hladnikovka (Hladnikia pastinacifolia Rchb.) je redka in potencialno ogrožena endemična vrsta, vključena v program krioprezervacije. Cilj diplomske naloge je določiti najbolj optimalno velikost vcepka, ki bi bila primerna za postopke krioprezervacije in najprimernejša gojišča, za regeneracijo takšnih vcepkov. Raziskali smo tudi učinek predpriprave pri 5 °C na regeneracijo vcepkov. Uporabili smo tri različne velikosti vcepkov brez predhodne krioprezervacije, in sicer majhne (0,5-1 mm), srednje (1-2 mm) in velike (2-3 mm). Le-te smo vcepili na tri različna gojišča, ki jih uporabljamo pri postopkih mikropropagacije, in sicer osnovno MS; osnovno MS z $2muM BAP$ (6-benzilamino purin); osnovno MS z $5muM BAP$ in $3muM IBA$ (indol-3-maslena kislina). Velikost vcepka je vplivala na preživetje in ponovno rast vcepkov. Najboljšo regeneracijo v brste smo dosegli pri velikih vcepkih na osnovnem gojišču MS, medtem ko smo najslabše rezultate dobili pri malih vcepkih. Vcepki na gojiščih z dodanimi rastnimi regulatorji so večinoma regenerirali v kalus ali propadli. Predpriprava je pozitivno vplivala na preživetje vcepkov, vendar je učinke predpriprave potrebno še dodatno raziskati.

Secondary title: The effect of explant size and growth media on regeneration of Hladnikia pastinacifolia Rchb. in tissue culture
Secondary abstract: Hladnikia pastinacifolia Rchb. is a rare and potentially endangered endemic species, involved in the program of cryopreservation. The purpose of the project is to investigate the influence of different culture media on the regeneration of different sizes of explants. Our goal is to determine the optimal size of the explant, which would still be appropriate for the procedures of cryopreservation. We are also investigating the most suitable culture medium for the regeneration of shoot tips. We have also studied the effects of pretreatment in a cool place at 5°C. We have used three different sizes of shoot tips: small (0,5-1 mm), medium (1-2 mm) and large (2-3 mm), which were moved to the different culture media, namely a basic MS medium; MS medium with added 2muM BAP (6-benzylaminopurine) and MS medium with added $5muM BAP$ and $3muM IBA$ (indole-3-butyric acid). The size of the explant is effecting on the survival and regrowth of them. The best survival and regrowth rate was achieved in large explants with basic MS medium, while small ones gave the worst results. Explants cultured on medium with added growth regulators usually regenerated callus or they died. Pretreatment had also a positive impact on survival of explants but it needs to be further investigated.
