Abstract
Namen diplomske naloge je določiti vpliv superkritičnega ogljikovega dioksida (SC CO2) na preživetje črnih kvasovk Trimmatostroma salinum in izločanje proteinov in nukleinskih kislin iz celične suspenzije T. salinum. Suspenzijo s kulturo T. salinum smo v visokotlačnem šaržnem reaktorju izpostavili SC CO2 pri tlaku 100 bar in 35 °C. Rezultati so pokazali, da se je s podaljševanjem časa inkubacije v SC CO2 preživelost celic T. salinum zmanjševala. Merili smo tudi aktivnost encimov celulaze, α-amilaze, β-glukozidaze in proteaze ter koncentracije proteinov in nukleinskih kislin v odvisnosti od inkubacijskega časa v SC CO2. T. salinum vsebuje ekstracelularne in intracelularne encime. Z inkubacijo celične suspenzije T. salinum v SC CO2 smo dosegli, da so se iz celic T. salinum izločili še intracelularni encimi. Tako se je posledično povečala koncentracija skupnih proteinov v suspenziji po inkubaciji v SC CO2. S podaljševanjem inkubacijskega časa celične suspenzije T. salinum v SC CO2 je padala aktivnost ekstracelularnim encimom, medtem ko se je aktivnost intracelularnih encimov v celični suspenziji T. salinum zviševala. Predvidevamo, da je SC CO2 povzročil denaturacijo encimov.

The purpose of the diploma thesis is to examine how the survival of black yeast Trimmatostroma salinum and secretion of proteins and nucleic acids are affected during exposure to supercritical carbon dioxide (SC CO2). The suspension of T. salinum was exposed to SC CO2 on high pressure batch reactor at 100 bar and at 35 °C. The results showed that the survival of T. salinum cells decreased with prolongation of incubation time in SC CO2. The activity of cellulose, α-amylose, β-glucosidase and protease in dependence of incubation time in SC CO2 was measured as well as the concentration of proteins and nucleic acids. T. salinum contains extracellular and intracellular proteins. During the incubation of T. salinum suspension in SC CO2, intracellular proteins were secreted. Therefore the total protein concentration in T. salinum suspension increased. With the increase in incubation time in SC CO2 the activity of extracellular enzymes in T. salinum cell suspension was reduced. We anticipate that the denaturation of enzymes was caused by SC CO2. |
