Abstract

Nikotinamid mononukleotid ali NMN je novost na trgu prehranskih dodatkov za potencialno zaviranje staranja. V številnih laboratorijih po svetu se ukvarjajo s študijami mehanizma delovanja v človeških celicah ter odkrivanjem blagodejnih in stranskih učinkov na organizem. Tedensko se objavljajo novi članki, ki ponujajo nove informacije o tem t.i. superživilu. Ni nujno, da je vsaka substanca, ki se pojavi na trgu, varna za uporabnike. Visoke cene novih superživil mnogokrat privlačijo tudi nepoštene proizvajalce zaradi želje po hitrem zaslužku. Zato je potrebno temeljito preverjanje kakovosti teh izdelkov. Namen naloge je bil razvoj HPLC metod za rutinsko določanje vsebnosti NMN in zaznavanje potencialnih sorodnih substanc v komercialno dostopnem izdelku. Razvoj aplikativne HPLC metode je zaradi posebne strukture NMN nekoliko zahtevnejši, vendar je zaradi enostavnosti in visoke razširjenosti uporaba tovrstne tehnike najbolj zaželena. V sklopu magistrskega dela smo uspešno razvili in validirali enostavni HPLC metodi z UV-Vis detekcijo za določanje vsebnosti NMN in njegovih sorodnih substanc v aktivni učinkovini.

Secondary abstract: Nicotinamide mononucleotide or NMN is the latest most interesting potential anti ageing food supplement. Numerous laboratories around the world are currently studying its mechanism in human cells, discovering benefits and side effects on human organism. New articles containing latest information are released on a weekly basis. However, not every substance brought to market is neccessarily safe for users. Unfair and untrustworthy manufacturers often take advantage of highly priced superfoods to obtain higher earnings. Thorough quality check of such products is therefore highly important. The purpose of this thesis was to develop an HPLC analytical method for routine determination of NMN assay and detection of potential related substances in commercial products. Due to NMN's unique structure, development of applicable HPLC method poses a bigger challenge. On the other hand, using an HPLC method is highly preffered due to its simplicity and global prevalence.
